Brain chemistry: the three systems, and what actually moves them
Dopamine drives pursuit. Serotonin holds your baseline. Norepinephrine sets how alert you are.*
That much is textbook neuroscience, and it is the part almost every article gets right. What almost none of them explain is that the three are not three separate dials. They share their raw material, they share their daily inputs, and one of them is built directly out of another.

Section 01
Two building blocks, three chemicals
Your body doesn't stockpile neurotransmitters. It builds them, constantly, out of amino acids you ate, and the assembly runs on ordinary nutrients you already know the names of.*
Dopamine and norepinephrine come from the same place. Both are built from tyrosine, an amino acid in almost any protein. Tyrosine becomes L-DOPA, L-DOPA becomes dopamine, and then a single further step converts dopamine into norepinephrine. The National Institutes of Health reference on catecholamines lays out that chain plainly, and the detail worth holding onto is the direction of it. Norepinephrine is downstream. It is made out of the dopamine you already have.
Serotonin arrives by a different road. Its raw material is tryptophan, a separate amino acid, and it travels a separate route with its own bottleneck and its own set of cofactors. That means two of your three run on one supply line and the third doesn't touch it.
The popular framing of three independent levers falls apart the moment you look at the plumbing. Push on the first and you have pushed on the second. Starve the shared input and two systems go quiet together.
The cofactors are the unglamorous part
Each conversion step needs help. Vitamin B6 shows up on both routes, iron on one, folate and magnesium alongside. None of it is exciting and all of it is required, because a pathway missing a cofactor is a pathway that runs slowly.*
This is the least marketable fact in the whole subject and one of the more useful ones. The machinery is nutritional before it is anything else.
Section 02
Levels are the wrong frame
Nobody can tell you your dopamine number. Not a clinic, not a mail-in kit, not an app. Urine and blood tests pick up what is circulating in the body, and that is largely a different pool from what is happening between two neurons behind your skull.
When a page offers to raise your serotonin level, it's describing something it can't measure and you can't check.
What researchers actually study is signaling. How much gets released, how long it stays in the gap between cells before it is pulled back, how sensitive the receiving side is that day. Those are the parts that change, and they change on a timescale of hours rather than months.
This is a system, not a tank. You're not filling something up, you're influencing how well a set of conversations runs.*
This is also the honest reason we do not publish a supplement that promises to raise a number. Nobody can verify that claim, including the person making it.
Section 03
What moves the system every day
The biggest levers here cost nothing, and they act on exactly the machinery section 01 just described. Get these right and everything else you do has a working foundation under it.
Sleep does real work here. Volkow used brain imaging in 2012 to look at what one night without sleep does to dopamine receptor availability, and it dropped measurably, tracking with how much alertness the participants had lost. That was total deprivation for a single night rather than a study of ordinary poor sleep, so it doesn't tell you what a bad fortnight does. What it does show is that the system responds to sleep quickly and visibly.
Movement and daylight both register. Morning light anchors the timing of the whole cycle, and exercise changes availability of the precursors and the sensitivity of the receiving side. Neither is a supplement story and both belong in any honest version of this one.*
Protein supplies the raw material. Tyrosine and tryptophan come from food, in ordinary amounts, from ordinary meals. Neither pathway needs an exotic input at the front of it.*
Read that list again and notice what it is. Four inputs, all of them within reach, all of them acting on the same machinery a supplement would act on. That's the optimistic version of this subject, and it happens to be the accurate one.

What the saffron research has actually tested
Saffron is one of the more heavily studied botanicals in this space, which is unusual for a spice and is why we built a product around it.
Jackson's team ran the trial that speaks most directly to an ordinary reader. 56 healthy adults, the kind carrying everyday stress rather than a diagnosis, took either standardized saffron or a placebo every day for 8 weeks.
At the end of it they all sat a lab stress test. The placebo group's heart rate variability fell under that pressure and the saffron group's held steady. That reading comes off the body rather than off a questionnaire, which is what makes it the most interesting result in the paper.*
The largest trial arrived in 2025, run by Lopresti and published in the Journal of Nutrition. 202 adults took either a standardized saffron extract or a placebo, at 28mg a day, for 12 weeks, double blind. It's the largest and best-controlled saffron trial anyone has run, and it's a good part of the reason this ingredient gets taken seriously. Its participants were recruited for low mood, so the place to read what it found is the paper itself.

Where it appears to act
Siddiqui reviewed the mechanism work in 2022 and the picture that comes out of it is a plant acting on the parts of the system section 02 described. Reuptake, the enzymes that break these chemicals down, and the signals that keep neurons responsive. Most of that evidence is laboratory work rather than human, which is normal for mechanism, and it did the job mechanism work is supposed to do. It pointed researchers at the systems the human trials then went and measured.*
The question people search most
More people ask about saffron and ADHD than about anything else to do with this plant, and the four trials run so far have come back on the encouraging side of the ledger.
The one to know is Pazoki's, because it is the only one that used a placebo and the only one in adults. 56 adults took saffron or a placebo alongside the medication they were already on, and after 6 weeks the saffron group came out ahead on one of the two symptom scales the researchers used.
The other three were in children and teenagers, and they compared saffron against methylphenidate instead of a placebo. In the head-to-head, both groups improved and the trial could not tell the two apart. In the two that added saffron on top of medication, the combination scored better than the medication alone.
These are small trials and the field is waiting on bigger ones. Saffron isn't an ADHD treatment, no regulator has approved it as one, and anyone weighing it for a diagnosis should raise it with the clinician managing that diagnosis.

Section 05
Three fair objections, answered straight
These are the arguments a careful reader makes, so they belong on our page rather than someone else's.
The studied dose is smaller than the one in the bottle
True, and worth being precise about. The trials keep landing on 28mg a day. Saffron360 has 88.5mg. You can do that division and some brands do, then present the ratio as though it were a multiple of the benefit, which the research does not support because the research validated the smaller number.
Our answer is that a higher dose is a formulation decision and not an efficacy claim, and we'd rather tell you the gap exists than hope you never do the arithmetic. More is not automatically better.*
Most of the mechanism work is not in people
Also true, and true of every field. Mechanism gets worked out in cells and animals first, because you can't open a living human brain to watch a reuptake transporter. The human trials then measure what people actually experience. Two different questions, answered by two different kinds of study, and saffron is one of the few botanicals in this category with a real body of both.*
Most of the research is in people who were already struggling
This is the objection that actually applies to you, and it's the one we'd raise if we were reading this page instead of writing it. Trials recruit people with a problem, because that's where a change is easiest to measure. Someone who already feels fine is a harder study to run and a much rarer one to fund.
Jackson's trial is the answer to that objection, and it's the reason it sits in section 04 rather than getting buried here. Those participants weren't patients. They were healthy adults carrying the ordinary load, which is most people reading this, and the stress-test result came out of that group rather than a clinical one. More trials in well populations would be welcome. This one already exists.*
- Standardization varies between extracts, so a milligram figure alone does not describe a reproducible ingredient.
- The largest trials used branded standardized extracts. That's ingredient-class evidence, not evidence about any one brand.
- Long-duration data is thin across this whole category, saffron included. The trials run weeks and months, not years.
Section 06
What you can actually verify
Here is the useful consequence of everything above. You cannot check a claim about your own neurotransmitters, and neither can the person selling to you. You can absolutely check what's inside the capsule.
That distinction matters more in this category than most, because saffron is the most expensive spice in the world and therefore worth faking. ConsumerLab bought 6 saffron supplements off the shelf in 2023 and tested them. 2 didn't contain the saffron compounds their labels described, and one of those showed a chemical pattern consistent with adulteration.
That leaves one question you can genuinely settle before you spend anything. Not whether a supplement raises a number, but whether the powder in the bottle is genuinely Crocus sativus, and whether the batch you're holding is the batch that was tested.
Ask for the certificate of analysis matching the lot number printed on your own bottle. A certificate with no lot number, or one that doesn't match, describes a batch you didn't buy.
